Fast Confirmation from First Growth Plate

  • Traditional culture confirmation requires not only the initial first growth plate, but subsequent growth plates on specific media.
  • Extra time consisting of many days may be needed to confirm the target microorganism.
     

 

 

MicroQuik unique enzyme detection systems consisting of a synthetic substrate which, in the presence of an enzyme specific to the infectious disease, causes a chemical reaction.

This reaction, when coupled to a developing agent, causes a color to appear on the filter membrane; or is visible under a fluorescent light without the addition of a developing agent.

   

A. Fluorochrome Detection - Detection requiring long wave UV light or reagent.  Results in 5 minutes.

B. Chromogenic Detection - Detection indicated by a color reaction.  Results in 30 minutes.

      1.  MicroQuik confirmation of E. coli
      2.  MicroQuik confirmation of Total Coliforms

C. Fluorogenic Detection - Detection requiring long wave UV light.  Results in 5 minutes.

      2.  MicroQuik confirmation of E. coli and Total Coliforms

 

1. Add 1 drop of Reagent A to the filter membrane.

2. Select colonies that morphologically resemble the target organism.
3. Touch the tops of 2-3 colonies with a loop, inoculating needle, swab, or wooden applicator.  Smear the colonies onto the filter membrane of the test card.


4. Set aside the inoculated card at room temperature for 5 minutes.
5. Add 1 drop of Reagent B to the filter membrane.  A purple color will immediately form on and around the deposited colonies in the presence of the hydrolyzed substrate, indicating a positive test. The test is negative if no purple color appears.  The test result must be read within 1 minute after the addition of Reagent B, otherwise the test result may be invalid.
   
    

 

1.  Add 1 drop of Reagent A to the filter membrane.
2.  Select colonies that morphologically resemble the target organism.
3.  Touch the tops of 2-3 colonies with a loop, inoculating needle, swab, or wooden applicator. Smear the colonies onto the filter membrane of the test card.


4.  Set aside the inoculated card at room temperature for 25 minutes.
5.  After 25 minutes, look for a blue/purple color around the smeared colonies. This is a positive test.  
     
The test is negative if no blue/purple color appears. A positive test is indicative of the target organism.

 

1.  Add 1 drop of Reagent A to the filter membrane.
2.  Select colonies that morphologically resemble the target organism.
3.  Touch the tops of 2-3 colonies with a loop, inoculating needle, swab, or wooden applicator. Smear the colonies onto the filter membrane of the test card.


4.  Set aside the inoculated card at room temperature for 5 minutes.
5.  After 5 minutes, look at the test card under a long wave (360nm) UV light. A positive test will be indicated by a blue/white fluorescence over a blue background around the smeared colonies.




                     
The test is negative if no blue/white fluorescence appears.  A positive test is indicative of the target organism.
